Abstract

Objective: GLP-1 is a key incretin hormone that not only regulates glucose metabolism but also contributes to energy expenditure and adipose tissue remodeling. However, the integrative role of GLP-1 in coordinating lipid oxidation and thermogenic gene expression in adipose tissues remains incompletely understood. Thus, we investigated anti-obesity effects of NEXITOP including regulation of incretin hormones and thermogenic genes related to fat metabolism. Materials and Methods: Male C57BL/6J mice were fed a 45% high-fat diet (HFD) for 14 weeks to induce obesity, followed by oral administration of NEXITOP, a novel GLP-1 regulator for 12 weeks. Serum incretin hormones markers (GLP-1, GLP-2, DPP-4) and glucose levels were analyzed, and expression of genes encoding thermogenic and fat oxidation markers (UCP1, PGC1α, ACOX1) was examined in white and brown adipose tissue. Results: NEXITOP administration significantly increased circulating GLP-1 and GLP-2 levels while reducing DPP-4 activity and fasting glucose. These hormonal changes were accompanied by a significant reduction in final body weight and visceral fat mass compared with the HFD group. At the tissue level, both white and brown adipose tissue exhibited upregulated expression of UCP1, PGC1α, and ACOX1, suggesting enhanced mitochondrial and peroxisomal fatty acid oxidation. Conclusion: NEXITOP ameliorates HFD-induced obesity by enhancing GLP-1-mediated signaling and reprogramming adipose tissue metabolism toward an oxidative and thermogenic phenotype. These findings highlight a potential therapeutic strategy combining incretin modulation and adipose metabolic remodeling for obesity and metabolic disorders.

Abstract

Objective: Chronic alcohol consumption causes neurotoxicity, yet moderate red wine consumption is linked to health benefits, often attributed to its polyphenols. This study investigated whether Nebbiolo red wine (NRW), a variety with high polyphenol content, possesses sufficient neuroprotective capacity to counteract the concurrent neurotoxic effects of its own ethanol (EtOH) content. Materials and Methods: Neuroprotective effects were evaluated in vitro (SH-SY5Y cells, WST-8 assay) and in vivo (mice, Y-maze test) following exposure to 14% EtOH or 14% NRW. Antioxidant capacity was determined by DPPH and ORAC assays. Results: NRW demonstrated robust dose-dependent antioxidant activity. In vitro, NRWP significantly protected SH-SY5Y cells from EtOH-induced cytotoxicity. In vivo, 14% EtOH administration severely impaired spatial memory (27.1% alternation vs. 67.5% Sham), an effect completely prevented by 14% NRW (65% alternation). Conclusion: NRW mediates significant neuroprotection, effectively neutralizing ethanol-induced cytotoxicity in vitro and cognitive impairment in vivo. This suggests polyphenolic content is a critical determinant of alcohol's net neurological impact.

Abstract

Objective: This study aimed to analyze teaching anxiety among Korean language teachers according to gender and native-speaker status, and to explore the characteristics of these anxiety experiences. Materials and Methods: A secondary analysis was conducted using factor analysis data from Chae & Cho (2025). One hundred in-service Korean language teachers participated, and teaching anxiety was measured using the Korean Anxiety Scale for Teachers (KLAST). Normality tests, independent samples t-tests, Welch t-tests, and Mann–Whitney U tests were conducted to compare anxiety levels by gender and native-speaker status. Results: Male teachers reported higher anxiety than female teachers regarding difficulties assessing student comprehension and meeting learner expectations. Non-native speaking teachers reported higher anxiety than native speakers concerning decreased student engagement, potential impacts of pronunciation or grammatical errors on lessons, and challenges managing classes due to weak student-teacher relationships. These results indicate that teacher anxiety is influenced by both individual and structural or situational factors. Conclusion: This study provides evidence that teaching anxiety among Korean language teachers varies by gender and native-speaker status. The findings highlight the need for tailored professional development and emotional support programs to enhance teacher well-being and educational practice.

Abstract

Objective: Eczema (Nār-Farsi), is a chronic, relapsing inflammatory skin disorder, significantly impairs patients' quality of life. In Unani system of medicine cause of Nār-Farsi is attributed to the excessive production of abnormal Sāfrā and Sāwda-e-Muhtariqā (yellow and burnt black bile). The treatment strategy primarily emphasizes Istifragh-e- Sāfrā and Sāwda (evacuation of yellow and black bile) and Ta‘dīl-i-Mizāj (moderation of temperament). Case report: This case report presents a 59-year-old female who attended the outpatient department (OPD) at the JJ Co-location Centre, Byculla -Mumbai, with complaints of persistent lesions on the right foot for the past two years. Symptoms included intense itching, skin roughness, pigmentation, and thickening. Based on clinical presentation, the condition was diagnosed as eczema resembling Nār-Farsi in the Unani system of medicine. The patient was managed with Unani formulations Itrīfal-i-Shāhtra, Sharbat-i‘Unnāb orally and Mārham-e-Kafoor topically. Treatment efficacy was evaluated using standardized tools, eczema area and severity index (EASI), patient-oriented eczema measure (POEM), and scoring atopic dermatitis (SCORAD), both before and after the intervention. Conclusion: This case demonstrated that Unani formulation effectively improves clinical symptoms such as itching, scaling, and pigmentation and may offer effective complementary management of eczema.
